Development And Research On Scanning-Laserbeam Bio-Sensing Microscope (In Japanese)

A novel scanning-laser-beam bio-sensing microscope, which can visualise the 2-dimensional distribution of pH value or concentration of a specific chemical substance in the specimen. Based on computer simulation results, the performance of the microscope was greatly improved: the spatial resolution higher than 10um, the precision of measurement higher than 0.01 in pH, and the measuring rate higher than 100 pixels/sec are achieved at present, and further improvement is in progress. This microscope can be applied to rapid and highly-sensitive observation of metabolic activities of microoorganisms. It was verified that growth of E.coli colonies on agar can be detected after 8 hours of incubation.
